Stefanini made her WTA Tour main-draw debut at the 2021 Abu Dhabi Open, qualifying for the main draw by defeating Irina Fetecău in the final qualifying round.

On 23 June 2008, she achieved her career-high singles ranking of world No. 35. She reached her first WTA Tour final at the 2008 Memphis Open but lost in straight sets to Lindsay Davenport.
